Using synthetic biology to harness O-GalNAc type glycosylation in Escherichia coli

Abstract
Human O-GalNAc linked glycans are responsible for a wide range of biological functions, and are often densely clustered on the carrier proteins, especially mucins. We are investigating the production and application of synthetic mucin derived glycoproteins. We are using engineered strains of E. coli to probe the use of mucin derived repeat sequences as means to extend the biological half-life of therapeutic proteins as well as learning how to make densely clustered uniform glycoforms to probe their potential biological activity.
